Samenvatting

GHRP-6 induces growth hormone release activity and affects the statuary growth in cattle. Use of this compound is prohibited in the European Union, in order to check this ban, methods have to be developed. The project involved developing a quantitative and confirmatory method for analysis of GHRP-6 in bovine urine and validation according to the European Commission directive 2002/657/EC. The sample clean-up involved using solid-phase extraction with Oasis HLB. Quantification of the compound was achieved by analysis with LC-MS/MS (QqQ) in positive mode. The sensitivity of the method was assessed by setting a validation level at 1 ng/mL and quantifying against matrix matched calibration range from 0.25 ng/mL to 5 ng/mL. Isotope dilution mass spectrometry was carried out to correct the results against matrix effects and ion suppression. Repeatability, reproducibility, accuracy, decision limit (CCα) and detection capability (CCβ) was used as validation parameters. The developed method proved to be specific, accurate (93%) and sensitive enough to detect the compound at 0.74 ng/mL (CCβ) and confirm at 0.37 ng/mL (CCα) with 3.4% measurement uncertainty.
